100,000 개의 단백질 structure 가 있다면 어떻게 나눌 수 있을까?

 > 비슷한 이름의 단백질은 거의 비슷한 구조를 지닌다. 그러므로 (교수님은) 4000개 정도일 것이라고 생각한다.

factors affecting protein..

1. rate of transcription of the gene.

2. efficiency of translation of mRNA into protein.

3. The rate of degradation of protein in the cell ( half-life ).

 

alpha - helix (first observed ), beta - sheet (second), random coil and turns (we don't know) , and another is almost same with Alpha and beta... so there isn't 3rd name..

 

Primary > Seconday > tertiary (3D, one single chain) > quaternary (interaction between different polypeptide unit .. )

N terminal (starting point), T terminal (end point)

sequence - structure relationship ( very important )...

Determining the protein structure/ polypeptide sequence by :

1. x-ray crystallography

2. Nuclear magnetic resonance

3. Protein predicting programmes - computer based (Bioinformatics)

 

Relationship between structure and function ( Important ) "생긴대로 논다... .."

A little part of proteins ( sometimes ) one to two function.. but normally one function has one structure..

 

Marker > 작은 부분으로 단백질의 일부분을 바꿀 수 있는 것... ex) Cancer Marker

In function prediction, the structure is quite important..

 

Modifications of protein..

Glycosylation/ Phosphorylation/ Sulfation > 단백질을 수정하여 문제가 일어날 수도 있고, 이를 이용하기도 한다.

 

SDS > point means a group (maybe almost same proteins)

Mass spectrometry

ex ) MALDI-TOF : MALDI > method of ionization, TOF > a mass analyzer

 

어떤 단백질이 세포 내에 존재한다는 것을 genome 만으로 증망할 수 없다.

Ligand > anything having relationship with proteins.
