Genes were introduced into HEK-293T or U2OS cells through transient transfection with Lipofectamine 2000 (Life Technologies). After 18–24 h (transfection), the medium was changed to 1 mL of fresh growth medium containing 250 μM desthiobiotin-phenol. This culture was incubated at 37°C under 5% CO2 for 30 min. Next, 100 μL of 10 mM H2O2 (diluted from 30% H2O2, H1009, Sigma Aldrich, St. Louis, MO, USA) was added to each well for a final concentration of 1 mM H2O2, and the plate gently agitated for 1 min at room temperature.
